1/0 or zero gauge wire is good and about 200 amp fuse near the battery. You don't need a fuse block at the rear of the vehicle unless you are splitting the power to multiple amps then fuse for the wire going to each amp.
With the upgraded wire you will be able to draw that much more from the charging system so you will likely have a higher voltage drop with the bigger wire. The only fix for this is to use a higher output alternator.
